L'engagement du récepteur d'antigčne des cellules T (TCR) est une étape clef dans l'activation et l'expansion des lymphocytes T (LT). En plus des signaux positifs de survie, de prolifération et de différenciation, le TCR peut induire des signaux négatifs d'anergie ou de mort. Nous avons étudié le signal de mort délivré par l'engagement du TCR dans les LT matures et le rôle de cette mort dans la régulation homéostatique du compartiment lymphocytaire T périphérique. Nous avons démontré que l'engagement d'un nombre supraoptimal de TCR induit une apoptose rapide des LT matures humains in vitro. Ce processus repose sur l'activation d'une nouvelle voie apoptotique impliquant le lysosome et le relargage cytosolique de cathepsines. Nous avons ensuite étudié la délétion précoce associée ŕ l'activation des LT in vivo. Suite ŕ l'injection du peptide spécifique ŕ des souris exprimant un TCR transgénique, nous avons observé une diminution précoce du nombre de LT activés par l'antigčne, qui correspond ŕ une apoptose et non ŕ une relocalisation de ces cellules. Cette apoptose est restreinte aux cellules activées par l'antigčne et n'est pas modifiée dans un contexte pro-inflammatoire.
